Can a Grimm actually forget the proclivities that make them a Grimm? I don't think so. If Nick is to be the Grimm poster boy, then series has gone through great effort to show that it cannot be done. Look at the way Nick lives and he might be considered more contemporary than most of them. Other than Monroe's birthday, when did we ever see him celebrate anything? Instead of being at the bar when he got his job back, he instead goes back into the workplace. Grimms memorialize themselves in books, revere their ancient weapons, and actively seek out the paranormal, the magical, and the mystical. Everything else comes in second. I think there's probably something chemical about them that gives them some kind of high when they see wesen experience fear of them. They adore the high their life gives them. What normal career or even a normal family life is ever going to compare to that?
If Marie were to be asked the question, would she have rather died quietly in bed as a librarian or in the hospital after a fight with wesen, how do you think she'd answer?
For that matter look at Nick's mother. She takes a hexenbiest to her son's house, and ends up losing her life fighting it out. I would imagine any minor regret she experienced at not raising her son didn't even compare to that last battle where she ends up losing her life.
Kelly did have regrets about leaving Nick which is why she advised him to live his life, with love and and all that because her choice to leave him meant a lifetime of regrets. However, living one's life, didn't take away what Grimms were destined to do (according to the finale) and that was to fight off evil, no matter what. Her dead spirit or whatever she was reminded him of his mission when he'd just about given up and was desperate to have his friends and family back. Not even cancer or a lifetime of retirement stopped Marie and Rolek from doing what was in their nature to do.
Kelly was killed because she was deceived into thinking her only son was in danger. I don't know any mother who loves their child who wouldn't return to help them when they needed it, even Renard's mother showed up a few times when he needed help.
