We don't even know the origin of Grimms and how they evolved to be what they are today. There was just some line in the finale about all of them being descended from the first Grimm who we have no idea how he/she came into existence.
Often when Grimm told you something it conflicted with another Grimm show. In one show, Nick tells Trubel that being a Grimm is a recessive trait and in another one Nick tells Trubel that at least one parent had to be a Grimm (dominant trait unique feature) for you to be a Grimm. A trait cannot be both recessive and dominant at the same time.
This is Nick talking and not a specific fact presented experientially on the show. He can be unclear because he's only representing his own opinion and not that of the show as a whole. Evidence would say that it definitely requires a Grimm parent. But it could also be recessive. We never meet any Grimm for whom this was the case, so it can't be verified. There is also a chance that this isn't entirely genetic. It's a possibility that there is some mystical component we're unaware of, limiting the number of Grimm capable of existing at any one time. But again, that's only speculative.
It is also possible something or several things might be needed to trigger it as well. The genes for some mental illness need a trigger which can be a virus when your mother was carrying you or can be caused by high levels of stress. Some people with the genes will never have mental illness.
It is possible that it takes some sort of trigger to activate the Grimm within. We know Trubel was assaulted, though I can't remember if she said whether or not that was the first Woge she ever saw. I'm not sure what it would have been for Nick. Maybe Adalind was so hot it blew his mind.
Maybe Sean had already hired her to hurt Maria, to get the key, because he knew Maria was dying and could be coming soon to Nick even though Nick did not know.
Women characters do not have to be having sex with the lead to be important to the story.
