I don't think it was ever said how Marie got the key. It could have been passed down in the Kessler family ever since the Crusades.
From what Renard told Nick, "they" would most likely have been the royals sending the verrat. I don't think we ever saw anyone other than grimms and royals who knew anything about the keys. In the absence of some history within the royal families that said they'd managed to get a key away from a Kessler, they'd naturally assume that Marie was the most likely keyholder and that she'd pass it to Nick, since they believed Nick's mom to be dead.
